摘要
以巨玫瑰葡萄花粉为试材,通过TTC法研究了不同发育阶段、干燥方式、贮藏条件、贮藏时间4个因素对巨玫瑰葡萄花粉生活力的影响,并采用培养基萌发法观察花粉萌发过程。结果表明:巨玫瑰葡萄盛花期花粉生活力(79.99%)明显高于初花期花粉生活力(62.59%)和末花期生活力(29.57%);室内自然阴干的花粉生活力(60.92%)平均高于干燥器干燥的花粉生活力(53.85%);常温、0℃、4℃及-18℃条件下贮藏的花粉平均生活力随温度降低呈升高趋势;花粉生活力随贮藏时间的增长逐渐减弱,-18℃贮藏30 d的花粉生活力可达26.55%。因此,盛花期花粉自然阴干是花粉的最佳采集方式,而长时间贮藏则以低温保存为宜。
The effects of development stage,desiccating methods,storage conditions and storage time on Jumeigui grape pollen viability were studied by TTC,and pollen germination process was observed in medium.The results showed that:grape pollen viability in flowering season (79.99 %) was higher than that at beginning of blooming period (62.59 %) and end of flowering (29.57 %).Average pollen viability by the shade drying (60.92 %) was higher than that by in desiocator drying (53.85 %).Average pollen viability incresed with temperature decreased in the condition of room temperature,0 ℃,4 and-18 ℃.The pollen viability stored in the condition in-18 ℃ for 30 days was 26.55 %.Therefore,the pollen in florescence should be placed in the shade to dry and the low temperature was the optimal storage conditions for long time.
