摘要
目的:建立测定人血浆中阿奇霉素浓度的方法并研究其药动学行为。方法:血样采用二氯甲烷处理后以高效液相色谱法进样测定。色谱柱为Kromasil C18,流动相为甲醇-乙腈-水-0.1 mol/L乙酸(39∶15∶46∶0.2),检测波长为210 nm,内标为罗红霉素。结果:阿奇霉素血药浓度在0.40~18.0μg/ml范围内线性关系良好(r=0.999 6);回收率〉80%,日内和日间RSD均〈10%。结论:本方法简便、快速、准确,适用于阿奇霉素药动学研究及血药浓度监测。
OBJECTIVE: To establish a method for the determination of azithromycin concentration in human plasma. METH- ODS: After treated with dichloromethane, the plasma sample was determined by HPLC. The determination was performed on Kro- masil Cls column with mobile phase consisted of methanol-acetonitrile-water-0.1 mol/L acetic acid (39:15:46:0.2). The detection wavelength was set at 210 nm, and roxithromycin was taken as the internal standard. RESULTS: The linear range of azithromycin was 0.40-18.0 μg/ml (r=0.999 6) ; average recovery was higher than 80%, and both intra-day RSD and inter-day RSD were be- low 10%. CONCLUSIONS: The method is convenient, rapid and accurate, and suitable for the pharmacokinetic study and plasma concentration monitoring of azithromycin.
