摘要
目的 分析甲状腺乳头状癌组(T组)及健康对照组(H组)血清代谢组学差异.方法 收集血清样本,采集图谱.利用Topspin、AMIX及SIMCA-P+软件进行图谱分析、数据积分并进行主成分分析及偏最小二乘判别分析,分析两组血清的内源性代谢差异.结果 以T组和H组数据建立判别模型,经交叉验证和排列实验验证,模型成立(P<0.05);T组中缬氨酸、亮氨酸、异亮氨酸、乳酸、丙氨酸、谷氨酸、赖氨酸、甘氨酸含量升高;脂类、胆碱、酪氨酸含量下降.结论 T组与H组之间存在明显的代谢差异.T组代谢异常不仅涉及糖代谢,而且涉及脂类、氨基酸及核酸代谢.
Objective Analysis the metabolic differences among the papillary thyroid carcinoma group (T group) and healthy controls group (H group) by nuclear magnetic resonance hydrogen spectrum.Methods Collecting the serum specimen,Analysis the serum specimen.To phase and baseline correction use Topspin software,to integral use AMIX software.Analysis the differences about endogenous metabolites between the T group and H group by principal component analysis (PCA) and parialleast square-discrimina (PLS-DA) with SIMCA-P + software.Results According to the data of T group and H group,discrimination model is established.By the PLS-DA cross validation and permutation test proving,the model is correct (P 〈 0.05).The content of metabolites in the serum of T group are increased including valine,leucine,isoleucine,lactic acid,alanine,glutamic acid,lysine,glycine,but the lipids,choline,tyrosine are reduced.Conclusion There is an obvious metabolic difference between T and H group.The difference is not only involve glucose metabolism but also the metabolism of lipid,amino acids and nucleic acid.
