摘要 目的:探讨新的自体表皮细胞悬液制取方法。方法:选取30只大鼠,应用切削皮法结合胰蛋白酶和组织微粒法结合中性蛋白酶/胰蛋白酶法两种技术获得自体表皮细胞悬液并进行细胞产量和活力的比较。结果实验组整个操作约1h内即可完成,而对照组则需要1.5天的时间。实验组活细胞率0.966±0.35,对照组活细胞率0.971±0.42,两组间比较无明显差异(P>0.05)。细胞产量:实验组(1.5±0.52)×106,对照组(1.9±0.72)×106,两组间比较有无差异(P>0.05)。结论:切削皮法结合胰蛋白酶法制取表皮细胞悬液,制取的表皮细胞悬液的产量和活力与组织微粒法结合中性蛋白酶/胰蛋白酶法比较无显著差异,但操作简单和耗时少。 Objective To investigate an improved method to prepare autologous epidermal cell suspensions. Methods 30 healthy wistar mice were choosed to prepare epidermal cell suspensions. An improved method including peeling grafts technique combined with trypsin was compared to atypical method including dispase and trypsin. Results It took one hour to prepare epidermal cell suspension with the improved method in test group.It took more than one day to prepare epidermal cell suspension with the typical method in control group.There were no significant differences for cell viability and cell yield of both groups (P〉O.05). Conclusion The improved method was a simple and effective method, it also took less time.
