云南省独龙族HLA-A,B,DRB1基因座多态性分析 被引量:1

摘要 目的研究云南省怒江州独龙江乡独龙族人群的HLA-A,B和DRB1基因座的等位基因及其组成的单倍型频率分布特征,为开展群体遗传学相关研究奠定基础。方法采用聚合酶链反应-直接测序分型法(polymerase chain reaction-sequence-based typing,PCR-SBT)对200名健康无血缘关系的个体进行HLA-A,B和DRB1基因座高分辨分型。结果200名云南省怒江州独龙族个体的HLA-A,B,DRB1基因座分别检出13、9、15个等位基因,其中HLA-A*02:01(55.75%),11:01(20%),HLA-B*56:01(65.5%),HLA-B*15:01(8%),HLA-DRB*114:01(55%),DRB*111:01(7.25%)基因频率较高。频率超过3%的单倍型分别为HLA-A*02:01-B*56:01-DRB*114:01(0.4277);HLA-A*11:01-B*56:01-DRB*114:01(0.0629);HLA-A*11:03-B*15:02-DRB*112:02(0.0450);HLA-A*02:01-B*38:02-DRB*108:03(0.0362);HLA-A*24:02-B*15:01-DRB*104:06(0.0343);HLA-A*31:01-B*35:01-DRB*114:03(0.0323)。结论云南省怒江州独龙族人群具有独特的HLA-A,B,DRB1基因频率分布特征,其HLA基因座的等位基因和单倍型具有较高的遗传多态性。HLA-A*02:01-B*56:01-DRB*114:01在该民族具有较高的频率。 Objective To investigate the genetic polymorphisms of HLA-A, B, DRB1 alleles and haplotype frequencies in Dulong ethnic group from Dulongjiang countryside of Nujiang county,Yunnan Province. Methods A PCR-SBT method was used to determine the alleles of HLA-A, B, DRB1 in 200 unrelated, healthy Dulong individuals. Results Altogether, 13 HLA-A alleles, 9 HLA-B alleles and 15 HLA-DRB1 alleles were observed in the population. HLA-A*02:01(55.75%),11:01(20%),HLA-B*56:01(65.5%),15:01(8%),HLA-DRB*114:01(55%),and 11:01(7.25%) were relatively high. 6 haplotypes frequencies of these loci were over 3%: HLA-A*02:01-B*56:01-DRB*114:01(0.4277);HLA-A*11:01-B*56:01-DRB*114:01(0.0629); HLA-A*11:03-B*15:02-DRB*112:02(0.0450);HLA-A*02:01-B*38:02-DRB *108:03(0.0362);HLA-A*24:02-B*15:01-DRB *104:06(0.0343);HLA-A*31:01-B*35:01-DRB*114:03(0.0323). Conclusion These results indicate that the HLA-A,B,DRB1 loci are highly polymorphic in Dulong ethnic group. HLA-A*02:01-B*56:01-DRB*114:01 are the most frequency haplotypein Dulongethnic group.
