摘要
目的:研究Beagle犬灌胃给予荷叶生物碱部位配伍何首乌二苯乙烯苷部位后荷叶碱的药动学特征,并与Beagle犬单独灌胃荷叶生物碱部位后荷叶碱的药动学参数进行对比,为荷叶制剂研制和临床应用提供实验依据。方法:Beagle犬灌胃给予荷叶生物碱部位0.15g/kg配伍何首乌二苯乙烯苷部位0.46g/kg后,定时取血,RPHPLC法测定血浆荷叶碱药物浓度,采用DAS软件计算药动学参数。结果:配伍何首乌二苯乙烯苷部位后,荷叶碱在Beagle犬体内药动学过程符合二室开放式模型;通过对配伍前后荷叶碱在Beagle犬体内的各药带动力学参数比较,结果表明,t1/2kα、t1/2α、Tmax和Cmax在两组中差异不大;配伍后荷叶碱的t1/2β(0.582h)小于单独灌胃荷叶生物碱部位后t1/2β(0.850h)。结论:配伍前后的药动学参数研究表明,配伍何首乌二苯乙烯苷部位后,不影响荷叶碱在Beagle犬体内的吸收、分布过程,但在一定程度上会加快荷叶碱在Beagle犬体内的消除。
Objective: To study on pharmacokinetic characteristics of nuciferine in Beagle dogs after the alkaloids fraction of Nelumbo nucifera. and the stilbene fraction of polygonum multil orum thunb were given in combination by gavage, and compared with the pharmacokinetic parameters obtained by the group which only alkaloids fraction of Nelumbo nucifera. were given by intragastric administration. To lay the foundation of herb compound research and set the experimental basis for its clinical application. Methods: The plasma samples were collected after intragastric administration of the alkaloid extract of Nelumbo nucifera 0.15g/kg combined stilbene fraction of Polygonum multil orum thunb. 0.46g/kg in Beagle dogs. The concentrations of nuciferine in plasma were measured by using RP-HPLC method and the pharmacokinetic parameters were calculated by DAS. Result: The concentration-time curves of nuciferine in plasma were fitted with a two-compartment open pharmacokinetic model in compatible group of Nelumbo nucifera. and Polygonum multil orum thunb.. There was no difference of t1/2kα, t1/2α, Cmax and tmax between the two groups. The pharmacokinetic parameter of t1/2β in compatible group was less than those only intragastric administration of the alkaloid extract of Nelumbo nucifera.. Conclusion: The pharmacokinetic parameters shows that, it had no effect on the absorption and distribution of nuciferine in Beagle dogs, but it can accelerate the elimination of nuciferine in Beagle dogs after compatibility.
