摘要
目的探讨交通心肾法对老龄雌性大鼠血管内皮L-精氨酸/一氧化氮合酶/一氧化氮(LArg/NOS/NO)通路的影响。方法选用15月龄雌性SD大鼠,随机将其分为老龄空白组、中药治疗组、西药对照组和中药对照组,分别给予生理盐水、交通心肾方药(20.65 g/kg)、戊酸雌二醇(0.12 mg/kg)和更年安(0.65 g/kg)灌胃,共28 d,另选3月龄大鼠作为青年组。采用放免法检测大鼠血清雌二醇(E2)、促卵泡刺激素(FSH)、促黄体生成素(LH),硝酸还原酶法测定主动脉外膜NO,比色法测定主动脉外膜NOS、L-Arg。结果与老龄空白组相比,中药治疗组NOS无明显差异(P>0.05),西药对照组和中药对照组NOS含量均明显下降,且有显著性差异(P<0.05)。与老龄空白组相比,中药治疗组和西药对照组L-Arg含量无显著性差异(P>0.05);中药对照组L-Arg含量明显下降,且有显著性差异(P<0.05)。与老龄空白组相比,中药治疗组和西药对照组NO均升高,且有显著性差异(P<0.05),中药对照组NO稍有升高,但无显著性差异(P>0.05)。雌激素和NO呈正相关性。结论交通心肾法能升高老龄雌性大鼠雌激素水平,对血管内皮L-Arg/NOS/NO通路有一定的调节作用。
Objective To investigate the influence of therapy of coordinating heart and kidney on vascular endothelial L-Arg/NOS/NO pathway in aged female rats.Methods Female SD rats (15 months old) were divided into aged group,Chinese medicinal group(20.65 g/kg),Western drug control group(0.12 mg/kg) and Chinese medicinal control group(0.65 g/kg),and respectively given orally normal saline (NS),Chinese medicinal of coordinating heart and kidney,Bujiale and Gengnian' an.Other rats (3 months old) were chosen as young group.The levels of serum estradiol (E2),follicular stimulating hormone (FSH) and luteinizing hormone (LH) were detected by using radioimmunoassay,level of aontic tunica.wdreritita nitric oxide (NO) was detected by using nitrate reductase method and levels of aortic tunica adventitia nitric oxide synthase (NOS) and L-Arg were detected by using colorimetric method.Results Compared with aged group,NOS level had no significant changes in Chinese medicinal group (P > 0.05),decrased significantly in Chinese medicinal control group and Western drug control group (P < 0.05).Compared with aged group,L-Arg level had no significant changes in Chinese medicinal group and Western drug control group (P > 0.05),decreased in Chinese medicinal control group (P < 0.05).Compared with aged group,NO level increased in Chinese medicinal group and Western drug control group (P < 0.05),and slightly increased in Chinese medicinal control group (P > 0.05).The level of estrogen was correlated to NO level.Conclusion The therapy of coordinating heart and kidney can improve estrogen level and has regulating effect on vascular endothelial L-Arg/NOS/NO pathway in aged female rats.
