摘要
目的:观察辛伐他汀对实验性大鼠脑出血(intracerebral hemorrhage,ICH)损伤中凋亡相关因子肿瘤坏死因子-α(TNF-α)表达的影响,探讨其通过抗凋亡机制发挥的脑保护作用。方法:采用注入新鲜自体不凝血复制脑出血模型,干湿重法检测脑组织含水量;免疫组化法检测TNF-α和caspase-3的表达,Western Blot检测脑组织TNF-α和caspase-3的蛋白表达水平。结果:大鼠脑出血后血肿周围基底节区脑组织TNF-α和caspase-3凋亡相关蛋白表达量、脑组织含水量及凋亡细胞数量明显上调。与模型组相比,辛伐他汀干预组的凋亡细胞数、TNF-α和caspase-3的表达与脑组织损伤周围含水量均显著降低(P<0.05)。结论:辛伐他汀对大鼠脑出血损伤有一定的保护作用,可能通过抑制脑组织中TNF-α和caspase-3的表达,进而减少细胞凋亡。
Objective: To observation of simvastatin on the expression of apoptosis-related factors of tumor necrosis fac-tor-α (TNF-α) in rat brain with intracerebral hemorrhage (ICH), and to study on the protective role played by the anti- apoptosis mechanism. Methods: ICH models was made by nonthrombogenic autolagous blood injection method, the water content of the brain was used to evaluate brain edema changes. Additionally, the expression of protein such as TNF-α and caspase-3 was determined by immunohistochemistry and Western Blot. Results: After cerebral hemorrhage, the number of apoptosis-related proteins of TNF-α and caspase-3 expression, the number of apoptotic cells and water content in brain tissue were significantly increased. The number of apoptotic cells , TNF-α and caspase-3 expression and damage the sur- rounding water content in brain tissue were significantly reduced in simvastatin intervention group compared with model group ( P 〈 0.05 ). Conclusion: Simvastatin is involved in certain protective effect of cerebral hemorrhage in rats, proba- bly by inhibiting TNF-α and caspase-3 expression in brain tissue, thereby reducing apoptosis.
