摘要
目的:探讨糖皮质激素地塞米松对大鼠脑出血后脑水肿的影响以及对TNF-α、TRADD、Caspase3、神经细胞凋亡的影响。方法:采用胶原酶注入法制作大鼠ICH模型;将制模成功的大鼠分为ICH组和地塞米松治疗组(治疗组),另设假手术组(对照组),每组36只;各组大鼠分别在造模后12h、1d、3d断头取脑组织检测脑水含量以及TNF-α、TRADD、Caspase3的蛋白表达,并进行TUNEL染色。结果:ICH组在12h、1d、3d脑水含量、TNF-α、TRADD、Caspase3蛋白表达以及凋亡指数均高于对照组(P<0.05);治疗组在上述时间点的各项指标均低于ICH组(P<0.05)。结论:地塞米松可降低ICH模型大鼠脑组织TNF-α、TRADD、Caspase3表达及减少神经细胞凋亡。
Objective: To study the effects of dexamethasone on the expression of TNF-α,TRADD,Caspase3 and on brain cells apoptosis after intracerebral hemorrhage in rats.Method: ICH model was produced in adult rats by infusion of collagenase Ⅶ into the caudate nucleus.The ICH model rats were randomly divided into dexamethasone treated group(treated group) and ICH group.And prepared other rats to be control group(36 rats in each group).The brain of rats were removed at 12 hours,1 day,3 days after operation.The brain water content and TNF-α,TRADD,Caspase3 expression were observed.Result: The brain water content and the expression of TNF-α,TRADD,Caspase3 and the apoptotic index were higher in ICH group than those of the control group(P〈0.05).These indicators were much lower than those of ICH group(P〈0.05).Conclusion: Dexamethasone can reduce the expression of TNF-α,TRADD,Caspase3,as well as neuronal apoptosis.
