类风湿关节炎中抗环瓜氨酸肽抗体、类风湿因子浓度与白介素17mRNA表达关系研究 被引量:8

Study on the relationship between serum anti-CCP antibody,RF and IL-17 expression in patients with rheumatoid arthritis

摘要 本文研究类风湿关节炎(rheumatoid arthritis,RA)患者血清抗环瓜氨酸肽抗体(anti-cyclic citrullinated peptide antibody,ACPA)、类风湿因子(rheumatoid factor,RF)浓度和白介素-17(interleukin 17,IL-17)mRNA表达水平的关系。收集40例RA患者血清和外周血,以40例健康对照者血清和外周血作对照。采用酶联免疫吸附法测定血清抗CCP抗体,免疫比浊法检测RF含量。Ficoll分离外周血单个核细胞,实时荧光定量PCR检测IL-17 mRNA表达。结果显示RA患者血清中ACPA和RF含量显著高于健康对照组(P<0.001);RA患者外周血单个核细胞中IL-17 mRNA表达量也显著高于健康对照组(P<0.01)。相关性分析显示RA患者血清中RF和ACPA含量与IL-17 mRNA表达呈正相关,相关系数分别为(RF,R^2=0.753;ACPA,R^2=0.2408)。提示在RA中,ACPA和RF含量与IL-17 mRNA表达具有正相关性,动态监测ACPA和RF含量及IL-17 mRNA表达,有助于RA的诊断和治疗。 To explore the relationship between serum anti-cyclic citrullinated peptide antibody(ACPA),rheumatoid factor(RF) and interleukin 17(IL-17) in patients with rheumatoid arthritis(RA),peripheral blood from 40 cases of RA and 40 cases of healthy donors(HD) were collected,and was subjected to isolation of serum and peripheral blood mononuclear cells(PBMC).Serum ACPA was detected by enzyme linked immunosorbent assay,RF was determined by immune turbidimetric and IL-17 mRNA expression was detected by real-time PCR.The results showed that both serum ACPA and RF in patients with RA were significantly higher than those of HD(P〈0.001).IL-17 mRNA expression in PBMC of RA was also significantly higher than that of HD.(P〈0.01).Analysis of serum levels of RF mRNA and IL-17 mRNA showed that they were significantly and positively correlated(R^2=0.753),while serum ACPA was positively correlated with IL-17 mRNA expression(R^2=0.2408).In conclusion,in RA,serum ACPA,RF and IL-17 mRNA expression are positively correlated.Dynamic monitoring of ACPA,RF and IL-17 mRNA was helpful to the diagnosis and treatment of RA.
