实时定量PCR检测类风湿关节炎滑液单个核细胞MIP-1α mRNA的表达 被引量:2

摘要 目的了解类风湿关节炎(RA)患者滑液单个核细胞MIP-1αmRNA表达情况。方法用实时定量PCR技术检测8例活动期RA患者滑液和血清单个核细胞(SFMC和PBMC)中巨嗜细胞炎症蛋白-1α(MIP-1α)mRNA的表达水平。同时检测血清CRP和滑液白细胞数。结果RA患者SFMC MIP-1αmRNA表达水平比自身PBMC表达水平明显增高(P<0.01),并且与血清CRP和滑液白细胞数呈正相关(r分别为0.9028和0.9120,P<0.01)。结论RA患者可能通过SFMC高表达MIP-1α趋化炎性细胞向滑膜迁移,加重局部炎症反应。 Objective To examine the expression of macrophage intlatrammtory protein-1 alpha (MIP-1α) by synovlal fluid monenuclear cells (SFMC) isolated from the synovial fluid (SF) of rheumatoid artlritis (RA) patients. Methods Real time polymerase chain reaction was used to determined the expression of MIP-1α mRNA by SFMC and peripheral blood mononudear cells (PBMC) isolated from 8 active RA patlents.The level of C reactive protein(CRP) in serum and the absolute white blood cell(WBC) count in synovial fluid were also analyzed.Results The mRNA level of MIP-1α in SFMC was significantly higher than that in PBMC (P 〈 0.01) and positively correlated with CRP( r = 0.9028, P 〈 0.01) and WBC eoant ( r = 0.9120, P 〈 0.01). Conclusion Overexpression of MIP-1α by SFMC of RA patients may contribute to the migration of inflammatory eells to the synovium and play a role in the pathogenesis of RA. reaction
