基于H5N1禽流感病毒HA2蛋白的通用型流感疫苗初步研究 被引量:3

Preliminary Study of a Universal Vaccine Based on the HA2 Protein of the H5N1 Influenza Virus

摘要 为研究有效的通用流感疫苗,解决流感疫苗株定期更换的难题。本课题利用乙肝病毒核心抗原(Hepatitis B core atigen,HBcAg)蛋白可以形成病毒样颗粒(Virus like larticle,VLP)的特点,将高致病性人禽流感A/Hubei/1/2010(H5N1)HA2的76~130氨基酸(Amino acid,AA)线性保守区(Linear conserved rgion,LCR)与HBcAg融合表达,并对其进行免疫学评价。其结果显示,优化后的LCR-HBc片段能够在pET30a大肠杆菌原核表达系统中大量表达。将纯化后的融合蛋白免疫小鼠,发现LCR-HBc疫苗组H5N1血凝素(Hemoglutination,HA)特异性IgG抗体滴度可达到1∶12 800以上。对免疫后小鼠的A/PR/8/34(PR8)致死剂量攻毒实验结果显示,LCR-HBc疫苗组肺病毒载量显著低于对照组(P〈0.01),对小鼠的保护率为50%。本研究的开展为流感通用型疫苗的研制提供科学线索。 Fragments encoding amino acids 76-130 in the linear conserved region (LCR) of A/Hubei/1/2010 (HSN1) HA2 was fused to hepatitis B core antigen (HBc) to generate a LCR-HBc virus-like particle (VLP). Results showed that the fusion protein of LCR-HBc was highly expressed in this prokaryotic ex- pression system. The purified LCR-HBc particle stimulated high levels of IgG production in mice with a ti- ter of 〉1:12 800, and provided 50% cross-protection against lethal challenge by HIN1 viruses.
