不同产地不同种质金银花中绿原酸和木犀草苷的含量分析 被引量:17

摘要 目的:建立金银花中绿原酸和木犀草苷含量的UPLC-PDA测定方法;在此基础上,以绿原酸和木犀草苷含量为指标,对9个产地金银花药材的质量及从山东曲阜引种到广西武鸣的2份种质两地间表现的品质进行评价。方法:采用Waters Acquity UPLC H-Class系统,Acquity UPLC BEH RP18(100 mm×2.1 mm,1.7μm)色谱柱,流动相为乙腈-0.4%的磷酸水溶液,流速为0.2 mL/min,梯度洗脱,检测波长242 nm。结果:山东曲阜所产的金银花药材品质较好,其绿原酸和木犀草苷含量分别为35.715和1.270 mg/g。广西武鸣从山东曲阜引种的九丰一号和树型金银花2种指标性成分含量均符合中国药典标准。结论:UPLC-PDA同时测定绿原酸和木犀草苷含量的方法简便、可靠,重复性好,可用于金银花质量评价;九丰一号和树型金银花可能成为广西武鸣的适应品种。 Objective: To establish an UPLC-PDA method for simultaneous determination of chlorogenic acid and luteoloside in Lonicerae Flos.On the basis of developed method,the quality of Lonicerae Flos from nine habitats and two local germplasms introduced from Qufu in Shandong to Wuming in Guangxi was evaluated.Methods: The analysis was performed on a Waters Acquity UPLC H-Class system.An Acquity UPLC BEH RP18( 100 mm × 2.1 mm,1.7 μm) column was used for all analyses.The investigated compounds were separated with a gradient mobile phase consisting of acetonitrile and 0.4% phosphoric acid solution at a flow rate of 0.2 mL /min,and the detection wavelength was set at 242 nm.Results: The quality of Lonicerae Flos from Qufu was the best among Lonicerae Flos of nine habitats for its content of chlorogenic acid and luteoloside at 35.715 and 1.270 mg /g,respectively.The content of chlorogenic acid and luteoloside in Lonicerae Flos of"Jiufengyihao"and "Shuxing"introduced from Qufu to Wuming both complied with the standard of Chinese Pharmacopoeia( 2010 edition).Conclusion: The developed UPLC-PDA method is simple,reliable and repeatable,which is helpful for the quality control of Lonicerae Flos."Jiufengyihao"and"Shuxing"are potential germplasms for the introduction of Lonicerae Flos in Wuming.
