摘要
目的 探讨碱性成纤维细胞生长因子 (basicfibroblastgrowthfactor,bFGF)和Ⅰ Ⅲ型胶原基因与视网膜激光损伤后修复的关系。 方法 用氪激光致伤有色家兔后极部视网膜 ,于伤后1,3,7,14d取眼球做冰冻切片 ,用组织原位杂交法观察bFGFmRNA和Ⅰ Ⅲ型胶原纤维mRNA表达的变化。 结果 光凝后 1~ 3d ,邻近光凝斑的兔视网膜节细胞层和内核层中bFGFmRNA阳性表达明显增加 ,在相同部位和时相Ⅰ Ⅲ型胶原纤维mRNA亦呈上调表达。 结论 Ⅰ Ⅲ型胶原基因与视网膜损伤修复直接相关 ,bFGF可能起重要的调节作用。
Objective To evaluate the relation of basic fibroblast growth factor (bFGF) gene and typeⅠ/Ⅲ collagen genes in the repair of rabbit's retina after laser photocoagulations. Methods The eyes of pigmented rabbit were injured at days 1,3,7 and 14 after the krypton laser photocoagulations. In situ hybridization was accessed to observe the expression changes of bFGF mRNA and collagen Ⅰ/Ⅲ mRNA. Results Significantly increased expressions of bFGF mRNA were found in the ganglion cell layer and inner nuclear layer adjacent to lesion region by 1 3 days after laser photocoagulations. The expressions of collagen Ⅰ/Ⅲ mRNA increased at the same site with time. Conclusions CollagenⅠ/Ⅲ mRNA directly relates to the repair of retina, bFGF may play an important regulative role.
