摘要
目的:观察Runt相关转录因子3(RunX3)、转化生长因子-β1(TGF-β1)及树突状细胞表面标志物CD83,在溃疡性结肠炎患者及正常对照组肠黏膜组织中的表达,探讨抑癌基因RunX3调节TGF-β1对树突状细胞在溃疡性结肠炎中的作用。方法:选取经过临床表现、内镜、病理等方法综合诊断为溃疡性结肠炎患者40例(UC组)及正常志愿者40例(对照组)结肠镜下乙状结肠肠黏膜活检标本,用Realtime PCR方法检测RunX3mRNA、TGF-β1mRNA和成熟树突状细胞常用的表面标志物CD83mRNA。结果:(1)与对照组肠道黏膜组织相比,UC组RunX3mRNA、TGF-β1mRNA的表达下调并有统计学差异(P<0.05),而CD83mRNA的表达上调且有统计学差异(P<0.05);(2)UC组肠黏膜组织中RunX3mRNA与TGF-β1 mRNA的表达呈正相关(r=0.629,P<0.05),RunX3mRNA与CD83mRNA的表达呈负相关(r=-0.456,P<0.05),TGF-β1mRNA与CD83 mRNA的表达呈负相关(r=-0.364,P<0.05)。结论:RunX3mRNA、TGF-β1mRNA及树突状细胞与溃疡性结肠炎密切相关,RunX3基因可能通过调节TGF-β1介导树突状细胞影响溃疡性结肠炎的发生和发展。
Objective:To observation expression of RunX3,TGF-β1and CD83(the marker of dendritic cells)in intestinal mucosa of patients with ulcerative colitis,and to investigate the effect of tumor suppressor gene RunX3 and TGF-β1on dendriti cell function in ulcerative colitis patients.Methods:A total of 40 ulcerative colitis patients confirmed by clinical manifestation,endoscopic,and pathological examination and 40 healthy volunteers were selected.Sigmoid colon mucosal biopsy specimens were detected by colonoscopy,RunX3 mRNA,TGF-β1mRNA and CD83 mRNA were determined by using the Real-time PCR method.Results:Compared with the control group,the expression of RunX3 mRNA,TGF-β1mRNA in patients with ulcerative colitis were lower than those in healthy controls(P〈0.05),and the expression of CD83 mRNA in patients with ulcerative colitis were higher than in those of healthy controls(P〈0.05).The expressions of RunX3 mRNA were positively correlated with TGF-β1 mRNA in ulcerative colitis patients(r=0.629,P〈0.05).The expressions of RunX3 mRNA was negatively correlated with CD83 mRNA in the ulcerative colitis patients(r=-0.456,P〈0.05).The expressions of TGF-β1mRNA was negatively correlated with CD83 mRNA in ulcerative colitis patients(r=-0.364,P〈0.05).Conclutions:RunX3mRNA,TGF-β1mRNA and dendritic cells are closely related with the ulcerative colitis.RunX3 gene may regulate TGF-β1-mediated dendriti cell function in ulcerative colitis patients.
