摘要
目的:探讨CCR9/CCL25通路对鼻咽癌CNE-2细胞增殖的影响。方法:培养CNE-2细胞,在培养体系中加入不同浓度的CCL25(0,10,25,50和100ng/ml),用MTT法检测各组细胞的增殖率,确定促CNE-2细胞增殖的最佳CCL25浓度。然后在培养体系中同时加入不同浓度的anti-CCR9(0,0.25,0.5,1,2μg/ml),观察此时CNE-2细胞的增殖情况。结果:不同浓度的CCL25均能促进CNE-2细胞的增殖(P<0.05)。而且其促细胞增殖作用呈现浓度依赖性,50ng/ml时达到最高水平。Anti-CCR9能阻断CCL25的促CNE-2增殖作用(P<0.05),而且也呈浓度依赖性。结论:CCL25具有促进CNE-2细胞增殖的作用,并呈现浓度依赖性。Anti-CCR9能阻断CCL25的促CNE-2增殖作用,并呈浓度依赖性。
Objective:To investigate the effect of CCL25 on the proliferation of NPC cell line CNE-2.Methods:For determining the optimized concentration of CCL25,we detected the proliferation rate in each subgroups of cultured CNE-2cells treated with CCL25.Then the proliferation status of CNE-2cells were observed after anti-CCR9 of different concentrations(0,0.25,0.5,1,and 2μg/ml)was simultaneously added into the culture systems.Results:All these studied concentrations of CCL25 could promote the CNE-2proliferations(P〈0.05).This promoting effect presented a feature of concentration-dependence and reached to the peak at 50ng/ml.Anti-CCR9 could block this promoting effect(P 0.05)also in a concentration-dependent manner.Conclusion:The CCL25 could facilitate the proliferation of CNE-2cells in a concentration-dependent manner.Anti-CCR9 could block the promoting effect of CCL25 on CNE-2cells and is also concentration dependent.
