摘要
目的建立高效液相色谱法实时监测患者血浆中茶碱的浓度,为临床合理用药提供参考。方法以Agilent ZorBax SB-C18柱(4.6mm×150 mm,5μm)为色谱柱,流动相为甲醇-1%乙酸铵(21∶79,v/v),流速:0.8 mL/min,检测波长272 nm,柱温:25℃,样品经20%高氯酸沉淀蛋白,高效液相色谱法测定茶碱血浆药物浓度。结果血浆中茶碱线性范围为2.62~31.44μg/mL,(r=0.999 7,n=5),检测限为0.5μg/mL。方法回收率为96.73%~104.71%,萃取回收率为90.46%~95.86%。日内日间相对标准偏差(RSD)〈3%。结论 HPLC法具灵敏,准确,快速,操作简便且经济等优点,适用于茶碱的血药浓度监测。
Objective To establish an HPLC method for the determination of theophylline in serum. Methods Separation and determination were achieved on a Agilent ZorBax SB-C18 column(4.6 ×150mm,5μm)with the mobile phase consisted of methanol-Ammonium acetate(21∶79)at a flow rate of 1.0 mL/min. The UV detection wavelength was set at 272 nm. The column temperature was 25℃. heplasma samples were treated with perchloric acid for sedimentation. Results The good linearity was from 2.62 to 31.44 μg/mL,(r =0.999 7,n =5). The minimum detectable concentration for theophylline was 0.5 μg/mL. The average recovery of relative and extraction was 96.73% ~104.71% and 90.46% ~95.86%. The within-day and between-day precision of variation was less than 3%. Conclusion The method of HPLC is accurate and reliable and suitable for clinical monitoring of theophylline.
