摘要
本研究采用液体发酵的方法,以发酵液活菌数为指标,采用单因素试验和正交试验对粪肠球菌HEW—A203的培养基和摇瓶发酵条件进行优化。确定最佳培养基的配方为:蔗糖3%,玉米淀粉4%,黄豆粉3%,磷酸氢二钾0.05%,硫酸镁0.1%,MnSO4·H2O 0.025%。最优发酵条件为:接种量2%、初始pH7.0、温度35℃、发酵时间10h、转速180r/min。采用该最优发酵工艺可以降低成本、提高质量,有利于粪肠球菌广泛应用于生产。
This study adopted liquid fermentation method, and use viable count of fermenta- broth as index, by the single factor and orthogonal experiment to optimize culture medium and fermentation conditions of HEW -A203 fermentation. Ultimately to determine the optimal medium composition: 3% sucrose, 4% corn starch, 3% soybean meal, 0.05% dipotassium hydrogen phosphate, 0. 1% magnesium sulfate; optimal fermentation condition is 2% inoculum size, initial pH 7. 0, temperature 35℃, fermentation time 10h, speed of revolution 180r/min. Optimal fer- mentation process could reduce costs, improve quality, and was more conducive to be widely used in production of Enterococcus faecalis.
