摘要
为了利用四氮唑脱氢酶还原法测定川贝母悬浮培养细胞的活力,并对影响其活力测定的各种因素进行了研究.选取四氮唑脱氢酶还原法测定了经三次继代培养的川贝母悬浮培养细胞的活力,并研究了不同四氮唑溶液浓度、缓冲液种类和pH值以及脱氢酶还原反应时间对川贝母悬浮培养细胞活力测定的影响.通过实验得出用四氮唑脱氢酶还原法测定川贝母悬浮细胞活力的最佳条件为:四氮唑溶液最适浓度0.6%,脱氢酶还原反应在Tris-HCl缓冲液中的效果优于磷酸缓冲液和磷酸氢二钠-柠檬酸缓冲液,最适pH为7.0 ~7.5;脱氢酶最适还原反应的时间为18 h.
The viability of frltillaria cell by suspension culture was measured by TrC dehydrogenase reduction method to study the effects of different tetrazole concentration,buffer typos,pH and the the reduction reaction time on the cell viability. The sults showed that the optimum conditions of the viability determination of fritillaria cell were that the optimum tetrazole concentration was 6 % ,the Tris-HC1 buffer was supe- rior to the phosphate buffer and disodium hydrogen phosphate-citrlc acid buffer,the optimum pH was 7.5 and the optimum reductlonreaction time was 18 h.
