中药蛇莓的薄层色谱鉴别及含量测定研究 被引量:5

摘要 目的:建立中药蛇莓的薄层鉴别及含量测定方法。方法:采用薄层色谱法为定性鉴别方法;用HPLC法测定蛇莓中鞣花酸的含量,色谱柱为汉邦ODS-3C_(18)柱(250mm×4.6mm,5μm),以乙腈-0.05%磷酸水为流动相进行梯度洗脱,流速为1.0mL·min^(-1),检测波长为254nm,柱温为30℃。结果:不同产地10批蛇莓药材的薄层色谱斑点清晰,且供试品色谱中,在与对照药材色谱相应的位置上,显相同颜色斑点;鞣花酸在进样量0.048~1.5μg范围内有良好的线性关系(r=0.9999),平均加样回收率为98.82%,RSD=3.14%(n=9)。结论:定性定量方法快速、准确、专属性强、重复性好,可用干蛇莓药材的质量控制。 Objective: To establish the methods for identification and content determination of Duchesneae Indicae Herba. Methods: TLC was used for the qualitative identification. The content of ellagic acid in Duchesneae Indicae Herba was determined by HPLC. The chromatographic separation was performed on the Hedera ODS-3 C18 column (250 mm × 4.6 mm,5μm) with acetonitrile-0.05% H3PO4 solution as the mobile phase, the flow rate was 1 mL·min^-1, and the column temperature was 30 ℃. The detection wavelength was 254 nm. Results: The TLC spots of 10 batch of Duchesneae Indicae Herba from different areas were clear. The spots in the chromatogram obtained with the test solution corresponded in position and color to those of the reference drug solution. The cali- bration curve of ellagic acid was linear in the range of 0. 048 - 1.5 μg ( r = 0. 9999) , and the average recovery rate was 98.82% with RSD of 3.14% (n = 9). Conclusion: The established qualitative and quantitative methods are rapid, accurate, specific and reproducible, and can be used for the quality control of Duchesneae Indicae Herba.
