摘要
目的探讨iNOS对兔角膜碱烧伤后新生血管形成的影响。方法选用健康大白兔30只建立角膜碱烧伤后新生血管模型,随机均分为两组。实验组、对照组分别予以选择性iNOS抑制剂氨基胍液与缓冲液滴右眼。观察并比较两组碱烧伤后第4天、第7天、第14天角膜新生血管生长情况及iNOS和VEGF蛋白的表达。结果实验组角膜新生血管面积、iNOS和VEGF蛋白的IOD值均较对照组显著性降低(均为P<0.05)。伤后第4天、第7天、第14天实验组和对照组角膜新生血管面积分别为(50.18±1.60)mm2、(86.53±2.27)mm2、(108.31±2.01)mm2和(70.04±2.85)mm2、(116.34±2.02)mm2、(151.40±0.58)mm2;伤后第4天、第7天、第14天实验组和对照组VEGF蛋白的IOD值分别为6410.51±156.16、4395.18±165.07、3566.27±236.48和13 092.53±159.42、10 682.30±253.65、8582.35±371.16;iNOS蛋白的IOD值分别为6566.09±194.56、4528.03±181.15、3472.62±131.87和12 618.60±196.63、10 438.68±176.89、8110.68±209.19。且iNOS和VEGF表达呈正相关(r=0.998,P<0.05)。结论 iNOS和VEGF可能参与了角膜新生血管的形成过程。氨基胍可通过选择性抑制iNOS的产生来降低VEGF的表达,进而抑制角膜新生血管的生长。
Objective To investigate the effects of iNOS on corneal neovascularization in rabbit after alkali burn. Methods Thirty healthy rabbits were selected to establish the model of corneal neovascularization after alkali burn and were randomly di- vided into two groups. Experimental group and control group respectively used aminoguanidine eye drops and buffer eye drops on right eyes. Corneal neovascularization and expression of iNOS and VEGF protein in two groups were compared on the 4th, 7th, 14th day after alkali burn. Results The corneal neovascularization areas and the IOD values of VEGF and iNOS in experimental group were significantly lower than those in control groups ( all P 〈 0.05 ). On the 4th,7th, and 14th day after injury ,the corneal neovascularization areas in experimental group and control group were (50. 18 ± 1.60 ) mm^2, ( 85.53 ± 2.27 ) mm^2, ( 108.31 ± 2.01 ) mm^2, respectively, and ( 70.04 ± 2.85 ) mm^2, ( 115.34 ± 2.02) mm2, ( 151.40 ±0.58 )mm^2, respectively. On the 4th, 7th, and 14th day after injury, the IOD values of VEGF in experimental group and control group were 6410.51 ± 156. 16,4395. 18 ± 165.07,3555. 27± 235.48, respectively, and 13092.53 ± 159. 42,10582.30 ±253.55,8582.35 ±371.15,respectively, and the IOD values of iNOS in experimental group and control group were 5555. 09 ± 194. 56, 4528. 0 ± 181. 15, 3472. 52 ±131.87, respectively, and 12518.60 ±195. 53,10438.58 ±175. 89,8110. 68 ±209. 19 ,respectively. At the same time,the expression of VEGF and iNOS was positively correlated ( r = 0.998 ,P 〈 0.05 ). Conclusion Both VEGF and iNOS may be participa-ted the process of corneal neovascularization. Aminoguanidine can degrade activity of VEGF through selectively inhibiting the expression of iNOS, accordingly suppressing corneal neovascularization.
