摘要
目的:探讨人脑胶质瘤组织中自噬相关基因Beclin-1的表达及与预后的关系。方法:采用免疫组化SP法及RT-PCR技术检测43例脑胶质瘤组织(包括Ⅰ级5例,Ⅱ级17例,Ⅲ级12例,Ⅳ级9例)及11例非瘤脑组织中Beclin-1蛋白和mRNA的表达,同时对随访资料进行Kaplan-Meier生存分析。结果:非瘤脑组织,Ⅰ级、Ⅱ级、Ⅲ级、Ⅳ级脑胶质瘤组织中Beclin-1蛋白和mRNA的表达水平差异均有统计学意义(F=99.962和41.951,P均<0.001)。Ⅲ级、Ⅳ级脑胶质瘤组织中Beclin-1蛋白和mRNA的表达水平低于Ⅰ级、Ⅱ级脑胶质瘤组织和非瘤脑组织(P<0.05),Ⅰ级、Ⅱ级脑胶质瘤组织中Beclin-1蛋白的表达水平低于非瘤脑组织(P<0.05)。43例患者随访到35例,以脑胶质瘤组织中Beclin-1蛋白表达水平的中位数为界限,将35例患者分成Beclin-1蛋白低表达组(16例)及高表达组(19例),两组的Kaplan-Meier生存曲线差异有统计学意义(χ2=4.962,P=0.026),Beclin-1蛋白高表达组预后优于低表达组。结论:Beclin-1可作为判断人脑胶质瘤预后的新指标用于临床。
Aim: To explore the relationship between the prognosis and the expression of autophagy-related gene Beclin-1 in human glioma tissue. Methods: Immunohistochemistry and RT-PCR technology were used to detect the expressions of Beclin-1 protein and mRNA in 43 cases of glioma tissue( including 5 cases of grade Ⅰ,17 cases of grade Ⅱ,12 cases of grade Ⅲ and 9 cases of grade Ⅳ) and 11 cases of non-tumor brain tissue. The follow-up data were analyzed by KaplanMeier survival analysis. Results: There were significant differences in the expressions of Beclin-1 protein and mRNA among non-tumor brain tissue,grade Ⅰ,Ⅱ,Ⅲ and Ⅳ glioma tissue( F = 99. 962 and 41. 951,P〈 0. 001). The expressions of Beclin-1 protein and mRNA in grade Ⅲ and grade Ⅳ glioma tissue were lower than those in grade Ⅰ,grade Ⅱglioma tissue and non-tumor brain tissue( P〈0.05). The expression of Beclin-1 protein in grade Ⅰ and grade Ⅱglioma tissue was lower than that in non-tumor brain tissue( P〈0.05). A total of 35 out of the 43 cases were followed up,16 cases with low expression of Beclin-1 protein and 19 with high expression based on the median expression level. The Kaplan-Meier survival curves of the 2 groups differed from each other( χ2= 4. 962,P = 0. 026),and the prognosis of high expression of Beclin-1protein group was better than that of the low expression group. Conclusion: Beclin-1 may be a new indicator to predict the prognosis of human glioma.
