长座虫草子实体和菌丝体中碱基和核苷类物质的HPLC分析 被引量:2

摘要 建立高效液相色谱法同时测定长座虫草中13种核苷和碱基类物质,色谱条件为:TSKgelODS-100V色谱柱(250mm×4.6mm,5斗m),以甲醇-0.Olmol/LKH2P04水溶液系统为流动相,梯度洗脱,流速1ml/min,检测波长260nm,柱温20℃。13种成分在50min内均达到基线分离,线性关系良好(r≥0.9990,n=8);平均回收率在98.4%~107.5%(RSD〈2.0%,n=5)。结果表明长座虫草子实体中胞嘧啶、次黄嘌呤含量相对较高;菌丝体中尿嘧啶、鸟苷、腺苷含量较高,尿苷在菌丝体和子实体中含量均较高。 Abstract An HPLC method was established for simultaneous determination of 13 nucleosides and nucleobases in Ophiocordyceps longissima. The separation was performed on a TSK gel ODS-100V column (250 mm x4. 6 mm,5 μm) using gradient elution with an aqueous potassium dihydrogen phosphate-methanol solution at 260 nm. The flow rate was 1.0 ml/min and column temperature was 20 ~C. Thirteen nucleosides and nucleobases were analyzed with- in 50 min. The calibration curves showed good linear relationship (r≥0. 999 0, n = 8 ) and the method exhibited excellent accuracy with a recovery of 98.4 to 107.5 % ( RSD 〈 2.0%, n = 5 ). Higher content of cytosine and hy- poxanthine were detected in the fruiting body. The contents of adenosine, uracil, guanosine were relatively rich in mycelium of Ophiocordyceps longissima. Uridine was abundant both in fruiting body and mycelium of Ophiocordyceps longissima.
