摘要
本研究以茄子叶为外植体,MS为基本培养基,研究激素NAA、ZT、6-BA不同浓度与组合对白撮茄子叶外植体分化的影响,并比较不同基因型外植体分化能力。结果表明:在不同培养基上白撮茄子叶均能诱导出愈伤组织,部分愈伤组织能分化形成不定芽,但出愈率、芽诱导率存在明显差别;诱导白撮茄愈伤组织分化最佳培养基为MS+NAA0.1mg/L+ZT3.0mg/L,出愈率为97%;诱导芽分化最佳培养基为MS+NAA0.1mg/L+ZT 4.0 mg/L+6-BA 1.5 mg/L+AgNO38.0 mg/L,出芽率为82%,AgNO3对外植体芽分化具有促进作用;白撮茄在1/2MS培养基中生根效果最好,生根率可达80%;不同基因型外植体出愈率、芽分化率有较大差异。本研究建立的高效稳定的茄子再生体系,为茄子遗传转化研究奠定了基础。
Using eggplant cotyledons as explants, different concentrations and combinations of hormones NAA, ZT,6-BA effects on callus induction were studied. Meanwhile, the explants differentiation of different genotypes was also discussed. The results shows that the Baicuo eggplant cotyledons could induce callus, partial callus can differentiate into adventitious bud, but induction rate was significant difference. The best inducing medium for Baicuo eggplant callus was MS+NAA 0.1 mg/L+ZT 3.0 mg/L and the callus rate was 97%. Best medium for inducing buds was MS+NAA 0.1 mg/L+ZT 4.0 mg/L+6-BA 1.5 mg/L+AgNO38.0 mg/L and the budding rate was 82%. AgNO3 has positive effect on bud differentiation. Best medium for rooting is 1/2MS and the rooting rate was 80%. The explants callus rate and bud differentiation rate are quite different among different genetypes. Baicuo regeneration system has been successfully established, which laid the foundation for the establishment of eggplant genetic transformation system.
