摘要 目的探讨盐霉素对人骨肉瘤MG-63细胞增殖凋亡的的影响及其分子机制.方法 MTT法检测盐霉素对人骨肉瘤MG-63细胞增殖的作用,利用流式细胞仪检测盐霉素对MG-63细胞凋亡的作用.Western blot法检测盐霉素对MG-63细胞中凋亡相关蛋白Bax、Bcl-2及Caspase-3蛋白表达的影响.结果盐霉素可以抑制人骨肉瘤MG-63细胞的增殖,并呈现出时间、剂量依赖效应;经过盐霉素(2μM和4μM)处理的MG-63细胞,与对照组(0μM)相比,其凋亡细胞的比率明显升高(P<0.05);盐霉素(2μM和4μM)还可明显上调促凋亡蛋白Bax、Caspase-3等表达水平,并同时下调抗凋亡蛋白Bcl-2的表达水平(P<0.05).结论盐霉素能有效抑制人骨肉瘤细胞MG-63的增殖,促进其凋亡,其作用机制可能与调节凋亡相关蛋白的表达有关. Objective To investigate the effect of salinomycin on the proliferation and apoptosis of human osteosarcoma cell line MG-63 and the underlying mechanism. Methods MTT assay was used to determine the effect of salinomycin on the proliferation of human osteosarcoma cell line MG-63. Flow cytometry was used to detect the apoptosis rate of MG-63 cell under salinomycin treatment. The protein expressions of apoptosis-related protein Bax, Bcl-2 and Caspase-3 in MG-63 cells under salinomycin treatment were evaluated by Western blot. Results The results showed that the treatment with salinomycin suppressed the growth of MG-63 cells in a time and concentration dependent manner; the apoptosis rate of MG-63 cells under salinomycin (2 μM, 4μM) treatment was remarkably increased as compared with that in the control group (0 μM) . In addition, Western blot results showed that the expressions of pro-apoptosis proteins Bax and Caspase-3 were significantly up-regulated under salinomycin (2 μM, 4 μM) treatment (P〈0.05) ; and the anti-apoptosis protein Bcl-2 was down-regulated (P〈0.05) Conclusion Salinomycin can inhibit the proliferation and induce apoptosis of human osteosarcoma cell line MG-63, which may be related to regulating the apoptosis-related protein expressions.
