摘要
目的观察盐霉素对乳腺癌细胞株MCF-7、MCF-7/ADR、MDA—MB-231的体外增殖和侵袭能力的影响,探讨其对乳腺癌细胞的转移抑制作用。方法用细胞计数试剂盒(CCK-8)法检测细胞增殖,得出半数抑制浓度(IC50)值;用Transwell小室法检测侵袭能力的变化;分别用实时定量聚合酶链反应(Real—timePCR)法和Westernblot法检测ETS1和基质金属蛋白酶(MMP)-1基因和蛋白的表达变化。结果盐霉素抑制乳腺癌细胞的增殖,3种细胞的IC50值分别为(7.47±0.64)、(5.11±0.97)、(4.08±0.85)μmoL/L;经不同浓度的盐霉素处理后,3种细胞侵袭能力逐渐下降,分别为对照组的65.9%/41.9%、55.8%/37.8%、48.5%/24.6%;MCF-7/ADR和MDA—MB-231的ETS1和MMP-1表达水平随着盐霉素浓度升高而下降。结论盐霉素可以抑制乳腺癌细胞的增殖,降低其侵袭能力。
Objective To study the effects of salinomycin on the proliferation and invasion of breast cancer cells and the mechanism. Methods The human breast cancer cells MCF-7, MCF-7/ADR and MDA-MB-231, were treated with salinomycin. Cell viability was determined by cell counting kit-8. The invasive ability of tumor cells was evaluated by Transwell chamber assay. The mRNA and protein levels of ETS1 and matrix metalloproteinase (MMP) -1 were detected by using real-time quantitative polymerase chain reaction (Real-time PCR) and Western blotting respectively. Results Salinomycin significantly in- hibited the proliferation of breast cancer cells with the IC50 value being (7.47 ±0. 64) for MCF-7, (5. 11 ±0. 97) for MCF-7/ADR, and (4. 08 ±0. 85) μmol/L for MDA-MB-231, respectively. The invasion capac- ity of breast cancer cells were significantly reduced by salinomycin (65.9%/41.9% for MCR-7, 55.8%/ 37.8% for MCR-7/ADR, and 48. 5%/24. 6% for MDA-MB-231, respectivel). Simustaneously, the ex- pression levels of ETS1 and MMP-1 were downregulated by salinomycin. Conclusion Salinomyein can in- hibit the proliferation of breast cancer cells, and reduce the invasion capacity.
