摘要
目的:通过研究不同时间点金欣口服液对呼吸道合胞病毒(RSV)感染模型A20表达的影响,进一步阐明金欣口服液治疗RSV肺炎的可能调节机制。方法:体外实验:RSV感染体外培养的RAW 264.7细胞,金欣口服液含药血清进行干预,24h后收集细胞,提取细胞蛋白,Western blot法检测各组A20含量;体内实验:RSV滴鼻感染BALB/c小鼠,金欣口服液不同剂量及给药方式进行干预,并于首次滴鼻后24、72、144h取各组小鼠肺组织,Western blot方法检测A20表达及金欣口服液的干预效果。结果:RSV感染RAW 264.7细胞及BALB/c小鼠后24、72、144h,A20表达均明显升高(P<0.01,P<0.05),而金欣口服液组较RSV感染组A20表达明显下调(P<0.01)。结论:金欣口服液能显著下调RSV感染模型后不同时间点肺组织A20的高表达,这可能是其治疗RSV肺炎的作用机制之一。
Objective: To further clarify the mechanism of regulation of respiratory syncytial virus (RSV) pneumonia by Jinxin Oral Liquid by study the effect of Jinxin Oral Liquid on the expression of A20 in RSV infected models at different time points. Methods: In vitro experiment: RAW 264.7 cells were infected with RSV, and treated with the drug serum of Jinxin Oral Liquid, cells were collected 24 hours later, cell protein was extracted to detect the content of A20 by using Western blot method. In vivo experiment: RSV was dropped nasally in BALB/c mice, and treated with Jinxin Oral Liquid of different dosage and administration methods. After 24, 72 and 144 hours of the first inoculated with RSV, the mice were sacrificed and lung tissues were collected to detect the expression of A20 by using Western blot method, and to observe the effect of Jinxin Oral Liquid intervention. Results: It demonstrated that infectin RAW 264.7 cell with RSV after 24, 72 and 144 hours of infection BALB/C mice with RSV, the expression of A20 was up-regulated significantly (P〈0.01, P〈0.05). The expression of A20 in Jinxin Oral Liquid group were down-regulated compared with that in RSV group (P〈0.01). Conclusion: Jinxin Oral Liquid can down-regulate the expression of A20 on lung tissues at different time paints of infection with RSV, which might be one of the mechanism for Jinxin Oral Liquid in treating RSV pneumonia.
