摘要
查尔酮合成酶是植物次生代谢途径中黄酮类物质合成的关键酶。使用同源基因克隆的方法,利用PCR技术从白芨中克隆到该类蛋白基因,命名为BsCHS(Genebank登陆号:KF812890),cDNA全长1 629bp,经DNAStar软件分析,包含一个完整的阅读框,编码390个氨基酸。不同组织的表达特征分析表明,BsCHS在花、茎中的表达较强,在叶和根部次之。蛋白质特征预测及蛋白序列结构分析发现BsCHS具有多个活性位点。序列同源比对表明该基因与蝴蝶兰、文心兰等的CHS基因有很高的相似性。
Flavonoids play an important physiological role in the growth and development of plants. Chalcone synthase is a key enzyme for flavonoid bio-sythesis in the plant secondary metabolite path- ways. In this study, the gene of BsCHS was cloned from Bletilla striata based on homologoμs genes from other species. RT-PCR analysis in different tissues indicated that BsCHS is expressed dominant- ly in flower and stem followed by leaf and root. The gene was 1 629 bp (KF812890) in length, enco- ding 390 amino acids. It is a protein with multiple active sites based on protein characteristics and se- quence structural prediction. Protein BsCHS was identified as a CHS gene according to its high iden- tity with Phalaenopsis sp. ' True Lady ' (accession : U88077) and Oncidiurn Gower Ramsey (accession : DQ118024).
