山猪MYF5基因SNPs及其形成的单倍型对部分肉质性状的影响 被引量:2

Effect of MYFS Gene SNPs and Haplotypes on Some Meat Quality Traits in Shanzhu

摘要 为了研究多个SNPs共同作用对肉质产生的作用,采用PCR-SSCP技术首次对杂种及纯种山猪的MYF5的全部外显子进行多态性分析,并利用SPSS statistics 16.0分析单个SNP及SNPs组成的单倍型对部分肉质性状的影响。结果表明:在第1个外显子内检测到2个SNPs,即955 bp处的A→C的突变,形成2种基因型(AA型和AC型),1 038 bp处的G→C的突变,形成2种基因型(GG型和GC型),2个突变位点均未发现纯合的突变型;A/C突变位点对蛋白质影响显著(P<0.05);A/C和G/C突变位点单独对剪切力的影响均不显著(P>0.05),组成的单倍型M-S(2个位点均突变)对剪切力影响显著(P<0.05),表明2个突变位点的互作会产生出新的遗传效应。3种单倍型AG、A-S和M-S在纯种山猪与杂种山猪中的分布存在明显差异;M-S单倍型不仅对蛋白质的影响显著(P<0.05),对剪切力的影响亦显著(P<0.05)。 In order to study the possible effects of multiple SNPs on meat quality, the PCR-SSCP was used to investigate different bloodlines of Shanzhu populations to find the polymorphisms of the total three exons of MYF5gene for the first time , and SPSS statistics 16.0 was used to analyze the association of MYF5 aflelic variants and haplotypes with some production traits. Result showed that a total of two SNPs on exon 1 were found. The first SNP was A→C in 955 bp which form two genotyoes(AA and AC); The second SNP was G--~C in 1 038 bpwhich form two genotypes (GG and GC), both of the mutations were not found homozygous mutant-type alleles. A/C allele significantly influence protein content (P〈0.05) but G/C allele don't( P〉0.05); Individual A/C or G/C allele had no significant effect on the shear force (P〉0.05), however, M-S type (both sites are mutated) had significant influence on shear force (P〈O.05), which mean interaction of two mutations will be generate new genetic effects. The distribution of three haplotypes A- G, A-S and M-S had significant difference in different bloodlines of Shanzhupopulations (P〈0.05); M-S type not only significantly influenced protein content but also shear force (P〈0.05). The study results provided an important theoretical reference for the molecular marker assisted breeding of pigs.
