肝舒胶囊抗鸭体内乙型肝炎病毒的实验研究

摘要 目的观察肝舒胶囊抗鸭体内乙型肝炎病毒(DHBV)的作用。方法以DHBV静脉注射感染雏鸭为模型,于感染后口服肝舒胶囊28 d,每日剂量分别为1.750、0.875、0.437 g·kg-1。采用斑点杂交方法检测鸭血清中的DHBV DNA;用ELISA快速法检测血清中鸭乙型肝炎病毒表面抗原(DHBSAG),检测用药前、后血清中DHBV DNA的滴度改变、DHBSAG的OD值改变情况。结果高、中剂量肝舒胶囊组用药后7~28 d均出现鸭血清中DHBV DNA的滴度明显降低,但在停药7 d后出现DHBV DNA滴度回升现象;肝舒胶囊3个剂量组治疗期间均对血清中DHBSAG有明显的降低作用。对DHBV DNA和DHBSAG的抑制作用均表现出一定的量效关系。结论肝舒胶囊对鸭体内DHBV有明显的抑制作用。 OBJECTIVE To observe the effect of Ganshu capsule on anti duck - hepatitis B virus (DHBV) in vivo. METHODS DHBV infected intravenously ducklings were used as a model. In 28 days after oral infection, Ganshu capsule was given at doses of 1. 750 g,0.875 g,0. 437 g·kg^-1. By dot hybridization detection, duck hepatitis B virus DNA (DHBV DNA)in serum was determined. Rapid ELISA assay was used measuring serum duck hepatitis B virus surface antigen (DHBsAg)in serum,Test before and after treat- ment DHBV DNA in serum titer of change, DHBsAg value changes of circumstances OD was also featured. RESULTS 7 to 28 days after treatment of high and medium dose, serum DHBV DNA titer was significantly reduced, but seven days after stopping rebound phenomenon occurs in DHBV DNA titer;During treatment of Ganshu capsule at three dose serum DHBsAg were significantly lowered. DHBsAg for DHBV DNA and inhibition showed a dose - effect relationship. CONCLUSION Ganshu capsule can significantly inhibit DHBV in liver in vivo.
