摘要
为了从八角叶片中提取高质量的基因组DNA,设计了7种DNA提取方法。结果表明,实验方法7提取的基因组DNA溶液无色透明,经0.8%琼脂糖凝胶电泳检测呈现出一条清晰的条带,蛋白、多糖、酚类物质等去除较彻底;PCR扩增及限制性内切酶消化证明,该DNA样品能够满足后续实验的要求。
Seven methods of DNA group extraction from Illicium verum were designed in order to obtain high quality DNA group. The result showed that the seventh method generated clear DNA solution which revealed clear strips when tested with 0.8% agarose gel electrophoresis and had protein, polysaccharide and phenolics deleted completely. PCR amplification and restrictive enzymes prove that the sample subject can meet the requirement of follow-up experiment.
