摘要
[目的]观察丹酚酸浸膏粉对气虚血瘀大鼠自由基代谢、血管内皮细胞分泌功能及相关生化指标的影响。[方法]用高脂饮食和游泳劳损及游泳后不予保温措施和擦拭水迹等方法致大鼠气虚血瘀模型,同时连续4W给予200mg·kg-1、100mg·kg-1、50mg·kg-1剂量的丹酚酸浸膏粉,观察对气虚血瘀大鼠自由基代谢、血管内皮细胞分泌功能、环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)、cAMP/cGMP等有关生化指标的影响。[结果]高、中、低剂量的丹酚酸浸膏粉可显著改善气虚血瘀大鼠出现的精神萎靡、四肢蜷缩、活动性差、毛发无光泽、粪便稀溏、舌质绛紫暗淡、眼球暗红、尾部皮下瘀紫等症状。连续给药4W后,气虚血瘀大鼠血清超氧化物歧化酶(SOD)活性明显提高(P<0.05,P<0.01),丙二醛(MDA)含量显著下降(P<0.05,P<0.01),血清一氧化氮(NO)、一氧化氮合酶(NOS)水平明显提高(P<0.05),血浆内皮素(ET)的含量显著下降(P<0.05,P<0.01),NO/ET比值明显升高(P<0.05,P<0.01),与阳性对照组1(丹参片)和阳性对照组2(地奥心血康胶囊)比,高、中剂量丹酚酸浸膏粉组的作用强于或相当于阳性对照1和阳性对照2,但差异均不显著(P>0.05);血糖(GLU)和甘油三脂(TG)显著降低(P<0.05);血浆环磷酸腺苷(cAMP)含量显著升高(P<0.05,P<0.01),血浆环磷酸鸟苷(cGMP)含量显著降低(P<0.05),cAMP/cGMP的比值明显提高(P<0.05,P<0.01),与阳性对照组比较,高、中剂量丹酚酸浸膏粉组的作用强于或相当于阳性对照1和阳性对照2,但差异均不显著(P>0.05)。[结论]丹酚酸浸膏粉能明显改善气虚血瘀大鼠的症状体征,提高气虚血瘀大鼠血清超氧化物歧化酶(SOD)活性和一氧化氮合酶(NOS)水平,降低丙二醛(MDA)和血浆内皮素(ET)的含量,对血管内皮有保护作用,从而改善患者预后。
[Objective] To observe the influence of Salvianolic acid extract powder on the free radical metabolism,secretion function of vascular endothelial cells and correlated biochemical index in qi deficient blood stasis rats.[Method] The level of SOD,MDA,NO,NOS,ET,GLU,TG,cAMP and cGMP in qi deficient and blood stasis rats induced by high fat diet and swimming strain were tested after treatment with 200mg·kg^-1,100mg·kg^-1,50mg·kg^-1Salvianolic acid extract powder for 4 weeks.[Result] Salvianolic acid extract powder could obviously improve the level of SOD（P〈0.01） and depress the level of MDA（P〈0.05,P〈0.01）.The contents of NO and NOS increased while ET decreased significantly（P〈0.05,P〈0.01） and the ratio of NO/ET was improved obviously（P〈0.05,P〈0.01）.The levels of GLU,TG and cGMP lowered while cAMP and cAMP/cGMP valued were improved（P〈0.05）.The effects of high and middle doses of Salvianolic acid extract powder were stronger than or equivalent to that of positive control 1 and 2 which had no significant difference（P〉0.05）.[Conclusions] The Salvianolic acid extract powder can improve the signs and symptoms of Qi deficiency and blood stasis rats,improve the SOD activty and NOS level,reduce the MDA and ET content,protect vascular endothelial and improve patients prognosis.
