摘要 目的观察IL-17对人牙周膜成纤维细胞RANKL表达的影响,探讨IL-17调控牙周炎骨改建的可能致病机制。方法用不同浓度(0、10、25、50 ng/mL)的IL-17处理人牙周膜成纤维细胞。选择最佳刺激浓度,用该浓度的IL-17处理细胞不同时间(0、12、24 h)。通过实时定量RT-PCR和Western-blot的方法检测细胞中骨吸收相关因子RANKL的表达水平。结果 IL-17可显著提高人牙周膜成纤维细胞RANKL的表达水平,并随着浓度的提高和刺激时间的延长而增加。结论IL-17可上调人牙周膜成纤维细胞的RANKL表达,提示IL-17可通过调控人牙周膜成纤维细胞骨吸收相关因子的表达参与牙槽骨的改建。 Objective To investigate the effect of interleukin^17 (IL-17) on the expression of RANKL and to explore the possible pathogenic mechanism of IL-17 regulating periodontitis bone reconstruction. Methods hPDLCs were treated with IL-17 of variable concentrations (0,10,25,50 ng/mL) ,and then the optimal concentration was used in the time-dependent experiment. The expressions of RANKL were assayed by real-time RT-PCR and western-blot. Results The expressions of RANKL were significantly up-regulated by IL-17 in a dose-dependent and time-dependent manner. Conclusions IL-17 can up-regulate the expression of RANKL of hPDLCs, suggesting that IL-17 can modulate the alveolar remodeling via regulating the expressions of pro-osteoclastie molecules of hPDLCs.
