烟草提取物对牙周炎小鼠Th17/Treg细胞免疫平衡及相关转录因子表达的影响 被引量:1

摘要 目的:探究烟草提取物(CSE)对牙周炎小鼠牙周组织的影响及与辅助性T细胞17(Th17)/调节性T细胞(Treg)免疫平衡的关系。方法:将小鼠随机分为Control组、Model组、CSE-L、CSE-M、CSE-H组;用牙周探针探查小鼠牙龈评估牙龈指数(GI);评估牙槽骨吸收;HE染色观察牙周组织病理形态学变化;流式细胞术检测牙周组织中Th17、Treg细胞比例;免疫印迹(WB)、免疫组化检测牙周组织白介素-17(IL-17)、视黄酸相关孤儿受体(RORγt)、IL-10、叉头状转录因子-3(Foxp3)蛋白表达。结果:与Control组相比,Model组GI、CEJ到ABC距离、Th17细胞比例、Th17/Treg比值升高,Treg细胞比例降低,IL-17、视黄酸相关孤儿受体(RORγt)蛋白表达升高,IL-10、Foxp3蛋白表达降低(P<0.05)。与Model组相比,CSE各组GI、CEJ到ABC距离、Th17细胞比例、Th17/Treg比值均进一步升高,Treg细胞比例进一步降低,IL-17、RORγt蛋白表达进一步升高,IL-10、Foxp3蛋白表达进一步降低(P<0.05)。结论:CSE可加重牙周炎小鼠牙周组织炎性反应、牙槽骨吸收,其机制可能与破坏Th17/Treg细胞免疫平衡有关。 Objective:To investigate the effect of cigarette smoke extraction(CSE)on periodontal tissues of periodontitis mice and its relationship with the immune balance of T helper cell 17(Th17)/regulatory T cell(Treg).Methods:Mice were randomly divided into Control group,Model group,CSE-L group,CSE-M group and CSE-H group;the gingival index was assessed by mouse gingival index(GI);evaluated the alveolar bone resorption;HE staining was used to observe the pathomorphological changes of periodontal tissue;the proportions of Th17 and Treg cells in periodontal tissues were detected by flow cytometry;the expressions of interleukin-17(IL-17),retinoic acid-related orphan receptor(RORγt),interleukin-10(IL-10)and fork-like transcription factor-3(Foxp3)in periodontal tissues were detected by Western blot(WB)and immunohistochemistry.Results:Compared with control group,the GI,CEJ to ABC distance,Th17 cell proportion and Th17/Treg ratio in Model group increased,the proportion of Treg cells decreased,the expressions of IL-17 and retinoic acid related orphan receptor(RORγt)increased,the expressions of IL-10 and Foxp3 decreased(P<0.05).Compared with Model group,the GI,CEJ to ABC distance,Th17 cell proportion and Th17/Treg ratio in CSE groups were further increased,the proportion of Treg cells decreased further,the expressions of IL-17 and RORγt increased further,the expressions of IL-10 and Foxp3 decreased further(P<0.05).Conclusion:CSE can aggravate periodontal inflammation and alveolar bone resorption in periodontitis mice,the mechanism may be related to the destruction of Th17/Treg cell immune balance.
