摘要
目的:建立蒲公英中槲皮素和木犀草素含量的测定方法.方法:采用反相高效液相色谱(RP-HPLC)法,色谱柱为Agilent HC-C18柱(250 mm × 4.6 mm,5μm);柱温:30℃;流动相:甲醇-0.4%磷酸(52∶48),流速:1.0 ml·min-1,检测波长:360 nm.结果:槲皮素和木犀草素的浓度分别在1.68~16.80 μg·ml-1(r =0.999 7),1.52~ 15.20 μg·ml-1 (r=0.999 8)时线性关系良好;平均加样回收率分别为98.67%和98.46%,RSD分别为2.49%和2.89%.结论:本方法测定快速,结果准确可靠,适合同时测定蒲公英中槲皮素和木犀草素的含量.
Objective:To establish the determination method for quercetin and luteolin in Taraxacum Herb.Methods:An Agilent HC-C18 column (250 mm× 4.6 mm,5 μm) was used at 30℃ with the mobile phase of methanol-0.4% phosphatic acid (52∶48).The flow rate was 1.0 ml · min-1.The detection wavelength was 360nm.Results:The linear range was within 1.68-16.80 μg · ml-1 for quercetin (r =0.999 7) and 1.52-15.20 μg · ml-1 for luteolin (r =0.999 8),the recovery of quercetin and luteolin was 98.67% (RSD =2.49%) and 98.46% (RSD =2.89%),respectively.Conclusion:The method is rapid,accurate and reliable,and suitable for the simultaneous determination of quercetin and luteolin in Taraxacum Herb.
