摘要 将致仔猪水肿病菌株107/86在TSB、TSA培养基上传代,使之表达F107菌毛,大量收集菌毛化的细菌,利用热洗脱法提取菌毛。粗提菌毛经15%的聚丙烯酰胺凝胶电泳可见一条约15ku的条带,以染色的凝胶为对照,将未染色凝胶上的菌毛条带切下,捣碎,加生理盐水,37℃,2h后离心,吸出上清再次电泳,可见提纯的菌毛条带。用针对F107菌毛的单抗做免疫印迹反应,证实此条带为F107菌毛蛋白。 Escherichia coli 107/86 which isolated from weaned pigs with edema disease was cultured in Trypticase soybroth. As shown by electron microscopy, the strain expressed fimbriae after growing at 37℃ for 24h.The bacteria werecollected and fimbriae were extracted by heat extraction(HE). Through SDS-PAGE, the crude fimbrial extract showed onemajor protein band of 15ku. Then the band was reextracted and purified from gels. Pure fimbriae were verified by Western-blot with monoclonal antibody (Mab) of F107.
