摘要
目的 探讨周期蛋白A(cyclinA)在原发性肝癌 (HCC)中的表达、临床意义及其与乙型肝炎病毒x基因整合之间的关系。方法 分别采用PCR、RT PCR、Westernblot检测HCC及其相邻非肿瘤组织中的cyclinA基因、mRNA和蛋白的表达 ;采用PCR和Southernblot检测HCC标本中HBx基因的整合。结果 35例HCC标本中 ,cyclinA基因扩增率为 2 .8% (1/ 35 ) ,mRNA过表达率为 4 5 .7%(16 / 35 ) ,蛋白过表达率为 6 0 .0 % (2 1/ 35 )。cyclinA蛋白过表达与患者年龄、肿瘤大小以及HBx基因整合之间存在统计学联系。结论 cyclinA的表达失控在肝细胞癌变的早期发生 ,可能是乙型肝炎病毒 (HBV)影响肝细胞正常细胞周期的重要途径之一。
Objective To investigate the frequency of cyclin A overexpression in hepatocelluar carcinoma(HCC) and its relationship with clinical significance an d HBx gene integration. Methods PCR?RT PCR and Western blot were used to detect the gene, mRNA and protein level of cyclin A in the tum or and nontumorous tissue. PCR and Southern blot were used to detect the integra tion of HBx gene in HCC. Results Amplification of cyclin A gene was found in 1 of 35 patients; overexpression of cyclin A mRNA and protein was found in 16 of 35, 21 of 35 patients, respectively . Overexpression of cyclin A protein was correlated with patient′s age?tumor s ize and HBx integration. Conclusion Overexpression of cyclin A occurs in the early stage of HCC carcinogenesis. It m ay be one of the important approaches by which HBV affects the normal cell cycle of hepatocyte.
