The Expression of Cyclin A,Cyclin D_1 and PCNA in Lung Cancer and Its Clinical Significance

摘要 目的观察细胞周期蛋白A、D1(Cyclin A、D1)和增殖细胞核抗原(PCNA)在肺癌中的表达及其临床意义。方法采用免疫组化和RT-PCR方法观察94例肺癌组织和10例假瘤炎性肺组织(对照组)中Cyclin A、D1和PC-NA的表达。结果在94例肺癌中,Cyclin A蛋白表达阳性率为84.0%;Cyclin D1蛋白表达阳性率为63.8%;PCNA蛋白表达阳性率为68.1%;肺癌的分期与Cyclin A和PCNA蛋白的表达相关(P<0.05)。在小细胞癌、鳞癌中Cyclin A、D1的mRNA表达较对照组均有非常显著的提高(P<0.01);在腺癌中Cyclin A、D1的mRNA表达较对照组也有明显增强(P<0.05)。结论肺癌中Cyclin A和PCNA蛋白表达有显著升高,Cyclin A和PCNA蛋白的过表达与肺癌的分期相关。 Objective To investigate the expression of Cyclin A,Cyclin D1 and PCNA in lung cancer and to determine its clinical significance. Methods 94 cases of lung cancer and 10 cases of normal lung tissues(control group) were observed by imrnunohistochemical SP method for the expression of Cyclin A,Cyclin D1 and PCNA. Results Of the 94 lung cancer cases,the positive expression rate of Cyclin A,Cyclin D1 and PCNA protein was 84.0%,63.8% and 68.1% respective- ly;Stages of lung cancer had a significant correlation with expression of Cyclin A and PCNA proteins(P〈0.05). The mRNA expression of Cyclin A,Cyclin D1 have highly significant raise comparison with control group in samll cell cancer and squamous carcinoma(P〈0.01);the mRNA expression of Cyclin A,Cyclin Dt have obviously enhancement facilitation comparison with control group in adenous cnacer(P〈0.05). Conclusion The expression of Cyclin A and PCNA proteins have highly significant raise in lung cancer, the over-expression of Cyclin A and PCNA proteins is related to the Stages and the poor prognosis of lung cancer patients.
