摘要
利用 RAPD技术 ,在花椰菜 ( Brassica oleracea ssp.botrytis)抗感黑腐病的近等位基因系之间进行 1 0碱基随机引物的筛选 .34 5条 RAPD引物扩增的产物表明 ,有 7条引物在近等基因系中呈多态性 .进一步的重复实验表明 ,只有引物 OP2 2 4能够产生稳定的多态性 .为了确定扩增产物 OP2 2 4/1 6 0 0与抗黑腐病基因的连锁程度 ,利用 F2分离群体进行检测 ,结果表明 :花椰菜种质 C71 2抗黑腐病生理种 BJ的抗性由一对显性基因控制 .RAPD标记OP2 2 4/1 6 0 0与抗病基因连锁 ,其遗传距离为 ( 4 .5± 1 .37) c
Three hundred and forty-five random sequence 10-mer primers were used to screen a pair of near-isogenic lines of cauliflower and their doner parent for markers linked to genes conferring resistance to Xanthomonas campestris pv. campestris (RXC). Seven primers were identified, which were seperated from the doner parent and the black rot resistance near-isogenic line but not from the recurrent parent. Of seven products that further investigated, only one reproducibly generated original product, and was confirmed by segregation analysis to be tightly linked to the RXC locus. The linked marker identified is useful for detecting the presence of the target gene in plant breeding and for a chromosome walk to cloning the gene.
