摘要
烟草 (Nicotiana tabacum L .)叶片在含有 2 mg/L 2 ,4 - D+0 .2 mg/L KT的 MS培养基上培养产生愈伤组织。用硫酸二乙酯 (DES)进行诱变处理 ,其半致死浓度为 0 .34%。处理后的愈伤组织接种到含有 15 %聚乙二醇 (PEG)的筛选培养基上 ,获得对水分胁迫具有抗性的愈伤组织 ,诱变率为 5 .4 %。抗性愈伤组织与对照相比 ,相对电导率和丙二醛 (MDA)
Callus of Tobacco(Nicotiana tabacum L.) was derived from leaves cultured on MS culture medium containing 2mg/L 2,4 D and 0.2 mg/L KT. Half death concentration of diethylsulfate (DES) which treatment for mutant inducement is 0.34%. When the treated callus inoculated on the selection medium contains 15% PEG, 5.4% of water stress resistant mutants were induced. Difference of the relative electrical conductivity and the content of malondialdehyde (MDA) between mutants and control reached very significant level.
