油菜细胞诱变及抗草酸突变体的筛选 被引量:10

摘要 以甘蓝型油菜品种中双一号愈伤组织为材料,用EMS、NaN3进行诱变处理.以草酸为选择剂进行了抗性筛选。经2%EMS处理的愈伤组织扩增3代后,从872块愈伤组织中.筛选到7块抗(耐)3.5mmol草酸的愈伤组织,其中有5块抗性稳定。抗性克隆的抗坏血酸氧化酶活性无甚变化,但多酚氧化酶以及过氧化物酶的酶活性都有显著的提高。 The calli cultures of Brassica napus cv. Zhongshuang No. 1 was treated with chemical mutagen and selection for oxalic acid resistant mutant was carried out. Results showed that suitable dosage of EMS (ethy1 methylene sulfonate) was 1 %~2% and the cultures was not sensitive to NaN3。From 872 clones asexually propagated 3 times from the callus treated with 2% EMS,7 clones resistant to 3. 5 mmol oxalic acid were screened out, and 5 out of 7 clones were stable in resistance. The activities of peroxidase and polyphenol oxidase were significantly higher than that of normal clones, but the activity of ascorbic acid oxidase did not change.
