三苯氧胺对肿瘤细胞的体外逆转耐药作用 被引量:8

In vitro experiment on reversal of multidrug resistance tumor cells by liophilic medicine tamoxifen

摘要 目的 探讨三苯氧胺 (tamoxifen ,TAM)逆转肿瘤细胞耐药性的可行性。 方法 以TAM联合化疗药物盐酸长春新碱 (VCR) ,羟基喜树碱 (HCPT)治疗人耐药肿瘤细胞KBv2 0 0 ,用四甲基偶氮唑盐比色法 (MTT) ,流式细胞仪PI染色法 ,荧光显微镜HT染色法对KBv2 0 0细胞行抑制率和凋亡率检测。 结果 单独应用 10 μg/ml的TAM无明显抑制肿瘤细胞的作用 ,而TAM联合化疗药物VCR、HCPT后 ,其抑制率及凋亡率均明显增加 (t =5 6 6 3,P <0 0 1;t =2 919,P <0 0 1)。 结论TAM能有效逆转肿瘤细胞KBv2 0 0对VCR 。 Objective To demonstrate the feasibility of tamoxifen (TAM) reversal of multidrug resistance (MDR) tumor cell line KBv200. Methods TAM was combined with VCR and HCPT chemotherapy in KBv200. MTT, PI coloration of flow cytometry and HT staining of fluorescence microscopy were performed to measure the inhibition rate and apoptotic rate of KBv200. Results Ten μg/ml TAM failed to inhibit tumor cell growth, whereas, when combined with VCR or HCPT, it inhibited tumor cell growth and caused cell apoptosis. The inhibition rate and the apoptotic rate were much higher than those of the VCR or HCPT alone. Conclusions TAM could reverse drug resistance of tumor cell KBv200, thus enhancing the chemosensitivity of tumor cells and providing evidence of reversal of MDR for clinical application.
