摘要
目的 探讨葡多酚 (GPC)对氧化损伤引发的细胞间黏附因子 1(ICAM 1)mRNA表达的影响作用。方法 人脐静脉血管内皮细胞株 (ECV3 0 4)在含有不同浓度GPC的培养液中培养 ,用Fenton体系产生的羟自由基 (·OH)造成细胞氧化损伤 ,反转录聚合酶链 (RT PCR)法检测各组细胞ICAM 1表达水平 ,并测定各组细胞内MDA含量。结果 高、中、低3个GPC剂量组的扩增产物电泳条带面积与阳性对照比较抑制度分别为 5 7 7%、5 1 1%、14 1%,中剂量组的MDA含量低于低剂量组。上述差异均有显著意义 (P <0 0 5 )。结论 GPC对氧化损伤引发的ECV3 0 4细胞ICAM 1mRNA表达有明显抑制作用。
Objectives The effects of grape procyanidins(GPC)isolated from grape seeds on expression of intracellular adhesion molecule-1(ICAM-1)mRNA induced by oxidative injury were investigated.Methods ICAM-1 mRNA expression of ECV 304 human umbilical vein vascular endothelial cells was detected with reverse transcriptase polymerse chain reaction (RT-PCR)after they were cultured with different doses of GPC and injured by hydroxyl radical (·OH) produced from Fenton reaction. Results Compared with positive control,areas of electrophoretic belts of the groups with higher dose GPC(100μg/ml)and lower dose GPC(10μg/ml)decreased by 51.1%and 17.2%,respectively(P<0.05).Conclusion GPC could resist expression of ECV 304 ICAM-1 mRNA induced by oxidative injury.
