摘要
本研究观察了对硫磷在体外条件下对大鼠脑突触小体钙稳态及cAMP量的影响。结果表明,对硫磷在较低浓度下即可明显抑制突触小体静息状态下的钙摄取功能,并使突触小体膜脂流动性下降。在较高浓度下,可抑制Ca^(2+)-Mg^(2+)-ATPase活性,增加cAMP含量。对硫磷的这些作用均存在剂量效应关系及时间效应关系,井与其抑制AchE活性的作用无关。
The activity of acetycholinesterase (AChE )and Ca2+-Mg2+ -ATRase, the amount of 45Ca uptake and of cAMP, and membrane lipid fluidity of rat brain synaptosome were measured in presence or absence of parathion. Marked inhibition of AChE activity ( 11-17% )and 45Ca uptake ( 13-42% )was observed in the presence of parathion at 2 -32×10-10mol/l and 5×10-9-5×10-7mol/l respectively. Reduction of Ca2+ -Mg2+ -ATPase activity (5-34% ) and elevation of cAMP level(130 -216% )were found with parathion at 6×10-5-5×10-8mol/l. Also, membrane lipid fluidity was decreased(8-16% )by parathion at 5×10-9-5×10-3mol/l. Based on the observations in vitro, we can see that the effects of parathion on calcium homeostasis and cAMP level were not related to the inhibition ot AChE activity. Besides, the results indicated that the non-cholinergic mechanisms of organophorsphorus may involve the change of calcium homeostasis and cAMP level.
