毒死蜱对大鼠脂质过氧化的影响 被引量:6

Effect of Chlorpyrifos on Lipid Peroxidation of Rats

摘要 目的探讨48%毒死蜱对大鼠胆碱酯酶活力及脂质过氧化作用的影响。方法将大鼠随机分为低、中、高3个剂量组和对照组,连续经口染毒28 d后,检测大鼠全血胆碱酯酶活力,血清及脑组织匀浆中丙二醛(MDA)含量、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-PX)活力。结果全血胆碱酯酶活力随染毒剂量的增加而降低,低剂量组、中剂量组、高剂量组胆碱酯酶活力相对值分别为87.9%(正常)、31.2%(中度中毒)、15.9%(重度中毒)。MDA含量在低剂量组升高,与对照组比较,差异有统计学意义(P<0.05),中剂量组、高剂量组降低。SOD(血清)、GSH-PX酶的活力随染毒剂量的增加而降低,各剂量组与对照组比较,差异有统计学意义(P<0.05),各剂量组组间比较,差异有统计学意义(P<0.05);脑组织匀浆中SOD活力随染毒剂量的增加而降低,各剂量组与对照组比较,差异有统计学意义(P<0.05)。结论毒死蜱在低剂量时可能主要通过脂质过氧化对机体造成损伤,在中、高剂量时可能主要通过抑制胆碱脂酶、SOD、GSH-PX的活力对机体造成损伤。 Objective To investigate the effect of 48% chlorpyrifos on cholinesterase activity and lipid peroxidation in rats.Methods Rats were randomly divided into low-,medium-,high-dose groups and control group.The rats were treated by oral gavage for 28 days.The activity of whole blood cholinesterase,the contents of malondialdehyde(MDA),the activities of superoxide dismutase(SOD)and glutathione peroxidase(GSH-PX)were detected in serum and brain tissue of rats.Results The activity of whole blood cholinesterase lowered with increased exposure,the relative values of cholinesterase activity of low-,medium-,and high-dose groups were 87.9%(normal),31.2%(moderate intoxication),and 15.9%(severe intoxication)respectively.The contents of MDA increased,the difference was statistically significant(P〈0.05)compared with the control group in the low dose group,and reduced in medium dose group and high-dose groups.The activities of SOD(serum)and GSH-PX lowered with increased exposure,the difference among various dose groups was statistically significant(P〈0.05)compared with the control group.The difference in-between various dose groups was statistically significant(P〈0.05).The activity of SOD in brain tissue lowered with increased exposure,the difference of various dose groups was statistically significant(P〈0.05)compared with the control group.Conclusions Chlorpyrifos injures the body probably through lipid peroxidation at low dosage,and in medium and high dosages probably by inhibiting the activities of cholinesterase,SOD,and GSH-PX.
