摘要
目的 研究 3种Bcl-2家族抗凋亡蛋白(Bcl-2、Bcl-xl、Mcl-l)在地塞米松(dexamethasone,DEX)诱导的不同类型人骨髓瘤细胞系凋亡过程中的作用。方法 采用MTF方法观察DEX对骨髓瘤细胞生长的影响;采用碘化丙腚(propidium iodide,PI)染色和流式细胞术分析骨髓瘤细胞经DEX处理后的凋亡情况;采用免疫印迹方法检测Bcl-2家族3种抗凋亡蛋白在骨髓瘤细胞凋亡过程中的表达改变情况。结果DEX能够诱导IL-6依赖型骨髓瘤细胞系XG-7和IL-6非依赖型骨髓瘤细胞系Sko-007发生典型的细胞凋亡反应;同时XG-7细胞中Bcl-xL和Mcl-1的表达在DEX处理后明显下调;而同样条件下Sko-007细胞中则出现了Bcl-2的降解。结论Bcl-2家族不同抗凋亡蛋白可分别介导DEX在不同骨髓瘤细胞系上的凋亡效应。
ve To investigate the function of three anti-apoptotic Bcl-2 family members( Bcl-2, Bel-XL and Mcl-l) in the apop-tosis of different human myeloma cell lines induced by dexamethasone (DEX). Methods Effect of DEX on the growth of myeloma cells was measured by MTT assay; Apoptosis of myeloma cells induced by DEX was determined by flow cytometry with propidium iodide (PI) staining of nuclei; The expression of three anti-apoptotic Bcl-2 family proteins in myeloma cells with or without DEX were determined by immunoblot assay. Results DEX induced a significant apoptosis of IL-6-dependent human myeloma cell line XG-7 and IL-6-independent Sko-007 myeloma cell line. In addition, Bcl-XL and Mcl-1 was down-regulated in XG-7 cells in the presence of DEX; while degradation of Bcl-2 can be observed in Sko-007 cells at the same conditions. Conclusion Different anti-apoptotic Bcl-2 family members can participate DEX-induced apoptosis of different human myeloma cell lines.
