摘要
为了获得细粒棘球绦虫 66ku抗原的重组蛋白和抗血清 ,用该抗原的cDNA克隆EgA3 1构建了pQE3 0EgA3 1重组质粒 ,在大肠埃希氏菌M15中表达 ,经亲和层析获得纯度较高的EgA3 1重组蛋白 ;用经复性处理的重组蛋白免疫动物 ,获得EgA3 1重组蛋白抗血清。本文就重组蛋白的稳定性和在复性处理中遇到的问题进行了讨论。
In order to prepare the recombinant protein of EgA31 cDNA representing the 66 ku antigen gene of Echinococcus granulosus and the antiserum, the recombinant plasmid pQE31 EgA31 was constructed and expressed in Escherichia coli M15. The recombinant protein was purified with affinity chromatography by means of using Ni NTA agarose. An anti serum against the recombinant protein was prepared throrgh animals being immunized with the recombinant protein which was purfied. The paper also discussed the stablity and the renaturation of the recombinant protein.
