摘要 聚合酶链式反应及单链构象多态性 (PCR-SSCP)技术作为一种区分检测基因组之间微小差异的有效方法 ,具有快速、简便、灵敏和适用于大样品量筛选的特点 .本研究从变性剂类型、电泳缓冲液的离子强度、交联度、电泳温度、甘油浓度及变性剂类型等方面对影响 PCR-SSCP的电泳条件进行分析 。 Polymerase chain reaction single strand conformation polymorphism (PCR SSCP) technique is a method feasible for detecting minor sequence changes in PCR amplified DNA, with its characterization of rapidness, simplicity, sensitivity and applicability to large scale screening. However, many factors can influence the results of SSCP analysis, and its optimization is highly empirical. In this paper, we compared and analyzed the experimental results under various controlled conditions such as the concentration of cross linker, the concentration of PAG, the concentration of the electrophoresis buffer, the presence of glycerol in gels and temperature of the gel during electrophoresis. Results showed that it was common to run the samples under at least two different conditions because each fragment needed its own optimal running conditions.
