摘要
为了摸索适宜于山茶属植物 DNA琼脂糖凝胶电泳条件 ,分析了电压、缓冲液、琼脂糖密度、EB( Ethyle Bromide)密度、染色时间及操作方法对山茶属植物 RAPD分析中琼脂糖凝胶电泳实验结果的影响 .结果表明 ,适宜于山茶属植物 DNA琼脂糖凝胶电泳条件是 :琼脂糖凝胶密度为 18g/L,电泳缓冲液为 TBE,电压为 6~ 9V/cm,电泳时间为溴酚兰和二甲苯青之间跑出约 5 cm,电泳开始时先让缓冲液与凝胶保持水平电泳 5 min,然后加缓冲液至浸没凝胶约 1mm深 ,继续电泳至结束 ,用 5 mg/L的 EB染色 40 min,水洗 40
The effects of agarose concentration,buffer, EB concentration,staining time and voltage imposed were studied .The results show that the optimal reaction condition is as follows:agarose concentration is 1.8%,TBE buffer,voltage imposed is 6~9 V/cm;at the beginning buffer should not immerse the gel,but just keep the same level of it.After 5 min,add some more buffer till it immerses the gel about 1 mm;in the end,stain the gel 40 min with EB and wash it 40 min with pure water.
