摘要
通过平皿计数法确定了嗜热链球菌ST、嗜酸乳杆菌C3和芽孢杆菌T12-1原生质体形成、再生和融合的最佳条件,以及筛选融合子的遗传标记物和选择培养基。在最佳条件下,ST、C3和T12-1的原生质体再生率分别为:10.5%、11.2%和8.6%;ST与T12-1的科间融合率约为4.6ⅹ10-6,C3与T12-1的科间融合率约为3.8ⅹ10-6 。比较了五种常用融合剂的促融效果,认为35% PEG6000促进原生质体融合的效果最好。
Optimums of formation, regeneration and fusion of protoplast from Streptococcus thermophilus ST, Lactobacillus acidlophilus C3 and Bacillus T12-1 were found by the method of plate counting. Heredity label objects of fusion hybrids and medium were prepared. Under these conditions, their regeneration rates were 10.5%、11.2% and 8.6% respectively; fusion rates of ST and T12-1, C3 and T12-1 were 4.6×10-6, 3.8×10-6 respectively. Otherwise, comparing the fused efficiency of five fusion agent, which was the best method that 35% PEG6000 promotes fusion.
